چکیده

Several transcription factors have been implicated as playing a role in myelopoiesis. PU.1, an ets-family transcription factor, is required for the development of myeloid and lymphoid lineages, whereas the transcription factor CCAAT–enhancer binding protein family member C/EBP is essential for granulocyte development. We present here the first evidence that C/EBP blocks the function of PU.1. PU.1 and C/EBP interact physically and colocalize in myeloid cells. As a consequence of this interaction, C/EBP can inhibit the function of PU.1 to activate a minimal promoter containing only PU.1 DNAbinding sites. We further demonstrate that the leucine zipper in the DNA-binding domain of C/EBP interacts with the 3/ 4 region in the DNA-binding domain of PU.1 and as a result displaces the PU.1 coactivator c-Jun. Finally, C/EBP blocks PU.1induced dendritic cell development from CD34 human cord blood cells. The functional blocking of PU.1 by C/EBP could be the mechanism by which C/EBP inhibits cell fates specified by PU.1 and directs cell development to the granulocyte lineage. Reciprocal Roles for CCAAT/Enhancer Binding Protein (C/EBP) and PU.1 Transcription Factors in Langerhans Cell Commitment

Myeloid progenitor cells give rise to a variety of progenies including dendritic cells. However, the mechanism controlling the diversification of myeloid progenitors into each progeny is largely unknown. PU.1 and CCAAT/enhancing binding protein (C/EBP) family transcription factors have been characterized as key regulators for the development and function of the myeloid system. However, the role...

PU.1 induces myeloid lineage commitment in multipotent hematopoietic progenitors.

Little is known about the transcription factors that mediate lineage commitment of multipotent hematopoietic precursors. One candidate is the Ets family transcription factor PU.1, which is expressed in myeloid and B cells and is required for the development of both these lineages. We show here that the factor specifically instructs transformed multipotent hematopoietic progenitors to differenti...

C/EBPalpha binds and activates the PU.1 distal enhancer to induce monocyte lineage commitment.

The PU.1 gene contains a 237-base pair distal enhancer located 14 kilobases upstream of its promoter. We have identified 2 sites within the PU.1 enhancer that strongly bind C/EBPalpha in a gel shift assay, and interaction with endogenous C/EBPalpha was confirmed by chromatin immunoprecipitation. Mutation of these DNA elements reduced activity of a distal enhancer-promoter construct 2- or 5-fold...
